United States. The QOS should reflect PSG expectations where they exist. For inhalation products, this usually means DDU and APSD methods, and may include spray pattern and plume geometry for sprays. Japan. Keep the QOS text simple and support it with clear cross-references. Where the Japanese method expectations differ from FDA PSG text, state the difference and justification in a few sentences and point to Module 3 for the evidence. Watch units and notation (for example, decimal separators) and keep naming exactly aligned with the Japanese sections. Do not change numbers across regions; change only the phrasing where required by local practice.

Process and Workflow: A Step-by-Step QOS Outline for Complex Generics

1) Product snapshot. One short paragraph that states the dosage form, route, strengths, pack, and device if present. Then list the key CQAs as bullet points. Keep it brief so a reviewer can see the scope without turning pages.

2) Control strategy table. A two-column table works well. Column one: CQA (for example, DDU, APSD fine particle fraction, IVRT release rate, Q3 rheology, particle size, release profile, dose accuracy). Column two: control measure (material control, in-process parameter, device specification, final test) with Module 3 pointers. This table should be consistent with Module 3 and should use the same attribute names.

3) In-vitro methods and acceptance criteria. For each method, state the purpose, the acceptance criterion, and the method capability in simple terms. Method capability means the method can detect meaningful change. A short sentence is enough: “The dissolution method detects a ±10% change in coating weight gain.” For topicals, state what Q3 attributes are compared and what ranges define sameness. 5) Specifications summary. Present a specification table that includes the test, method (with ID), acceptance criterion, and the Module 3 location. Keep numbers identical to Module 3. Include performance-relevant tests (for example, release rate, IVRT, APSD) in the same table or in a second table if needed. 6) Method validation summary. Keep the QOS concise.
